Neom sounds like something straight out of a science-fiction novel.

Buried in the 2,300 pages of planning documents obtained by the WSJ are plans for the forcible relocation of local tribes, the creation of human gene-editing clinics, and 24/7 government surveillance — meaning this futuristic city may be more like dystopian sci-fi than the fun Jetsons kind. Since 2017, Saudi Crown Prince Mohammed bin Salman, better known as MBS, has been working with teams of consultants to develop his vision 2030 for the $500 billion city in the currently barren northwestern corner of Saudi Arabia.

It’d be easy to get distracted by all the shiny things detailed in the Neom planning documents — but the futuristic city also sounds suspiciously like a totalitarian surveillance state. “This should be an automated city where we can watch everything,” Neom’s founding board is quoted as saying in the documents, according to the WSJ. “[A city] where a computer can notify crimes without having to report them or where all citizens can be tracked.”

That means while you’re chilling on the glowing beach, daydreaming about your next prix fixe meal, a drone equipped with facial-recognition technology will likely be transmitting your location to Neom’s “1984”-esque law enforcement officials. That’ll be thanks to all the medical clinics genetically modifying Neom’s citizens to be stronger, not to mention smarter — meaning MBS might not just be planning to create the city of the future, but the humans of it as well.

New research has revealed that it may one day be possible for a male couple to create a biological child together.

Scientists at Oregon Health & Science University (OHSU) have been working on a process called in vitro gametogenesis (IVG), in which they create an egg from a skin cell and then fertilize it with sperm.

The coverage will also extend to single folks and will finally allow for the use of donated eggs, sperm, and embryos.

The researchers have been using mice for the process, which involves removing the nucleus of a skin cell and transferring it into a donated egg that has had its nucleus removed. From there, the scientists eliminate half the chromosomes in the nucleus so a sperm cell can fertilize it. A similar process was used to clone Dolly the sheep in 1996, though that did not involve removing and replacing half the chromosomes.

The study’s senior author, Shoukhrat Mitalipov, director of the OHSU Center for Embryonic Cell and Gene Therapy, and professor of obstetrics and gynecology, and molecular and cellular biosciences, in the OHSU School of Medicine, told OHSU News that the goal “is to produce eggs for patients who don’t have their own eggs.” IVG could potentially be used to assist those with uteruses experiencing infertility as well as male same-sex couples in producing biological children.The scientists are reportedly studying how it could work with human cells, and while there have been key developments, it will still be years before anything is put into practice.

“This gives us a lot of insight,” said Paula Amato, professor of obstetrics and gynecology at OHSU School of Medicine. “But there is still a lot of work that needs to be done to understand how these chromosomes pair and how they faithfully divide to actually reproduce what happens in nature.”

As of now, the chromosomes have only paired correctly in rare cases, so the next step of the research is to “determine how we enhance that pairing so each chromosome pair separates correctly,” explained Mitalipov. This study is considered unique from the way other researchers have approached IVG. Many scientists are working on a process that involves reprogramming skin cells into stem cells and then using them to create egg or sperm cells.

“We’re skipping that whole step of cell reprogramming,” said Amato. “The advantage of our technique is that it avoids the long culture time it takes to reprogram the cell. Over several months, a lot of harmful genetic and epigenetic changes can happen.”

Human artificial chromosomes (HACs) capable of working within human cells could power advanced gene therapies, including those addressing some cancers, along with many laboratory applications, though serious technical obstacles have hindered their development. Now a team led by researchers at the Perelman School of Medicine at the University of Pennsylvania has made a significant breakthrough in this field that effectively bypasses a common stumbling block.

In a study published in Science, the researchers explained how they devised an efficient technique for making HACs from single, long constructs of designer DNA. Prior methods for making HACs have been limited by the fact that the DNA constructs used to make them tend to join together—”multimerize”—in unpredictably long series and with unpredictable rearrangements.

The new method allows HACs to be crafted more quickly and precisely, which in turn will directly speed up the rate at which DNA research can be done. In time, with an effective delivery system, this technique could lead to better engineered cell therapies for diseases like cancer.

“Essentially, we did a complete overhaul of the old approach to HAC design and delivery,” said Ben Black, Ph.D., the Eldridge Reeves Johnson Foundation Professor of Biochemistry and Biophysics at Penn. “The HAC we built is very attractive for eventual deployment in biotechnology applications, for instance, where large scale genetic engineering of cells is desired. A bonus is that they exist alongside natural chromosomes without having to alter the natural chromosomes in the cell.”

The first HACs were developed 25 years ago, and artificial chromosome technology is already well advanced for the smaller, simpler chromosomes of lower organisms such as bacteria and yeast. Human chromosomes are another matter, due largely to their greater sizes and more complex centromeres, the central region where X-shaped chromosomes’ arms are joined.

Researchers have been able to get small artificial human chromosomes to form from self-linking lengths of DNA added to cells, but these lengths of DNA multimerize with unpredictable organizations and copy numbers—complicating their therapeutic or scientific use—and the resulting HACs sometimes even end up incorporating bits of natural chromosomes from their host cells, making edits to them unreliable.

In their study, the Penn Medicine researchers devised improved HACs with multiple innovations: These included larger initial DNA constructs containing larger and more complex centromeres, which allow HACs to form from single copies of these constructs. For delivery to cells, they used a yeast-cell-based delivery system capable of carrying larger cargoes.

“Instead of trying to inhibit multimerization, for example, we just bypassed the problem by increasing the size of the input DNA construct so that it naturally tended to remain in predictable single-copy form,” said Black. The researchers showed that their method was much more efficient at forming viable HACs compared to standard methods, and yielded HACs that could reproduce themselves during cell division.

The potential advantages of artificial chromosomes—assuming they can be delivered easily to cells and operate like natural chromosomes—are many. They would offer safer, more productive, and more durable platforms for expressing therapeutic genes, in contrast to virus-based gene-delivery systems which can trigger immune reactions and involve harmful viral insertion into natural chromosomes.

Normal gene expression in cells also requires many local and distant regulatory factors, which are virtually impossible to reproduce artificially outside of a chromosome-like context. Moreover, artificial chromosomes, unlike relatively cramped viral vectors, would permit the expression of large, cooperative ensembles of genes, for example to construct complex protein machines. Black expects that the same broad approach his group took in this study will be useful in making artificial chromosomes for other higher organisms, including plants for agricultural applications such as pest-resistant, high-yield crops.

Researchers from the J. Craig Venter Institute, the University of Edinburgh, and the Technical University Darmstadt were also involved in the study.

DNA, the molecule that stores the genetic information of all living things, is made up of just four chemical letters, or nucleotides. But what if we could add more letters to this alphabet and create new kinds of DNA?

That’s what a team of researchers from the University of California San Diego, the Foundation for Applied Molecular Evolution, and the Salk Institute for Biological Studies have done. They have developed a new version of DNA with six letters instead of four, showing that it can be used to make proteins, the building blocks of life. This feat, published in Nature Communications, opens doors to a future where custom-designed proteins and novel biological applications could become a reality.

Four nucleotides

DNA, the blueprint of life, encodes its instructions using just four nucleotides – adenine (A), thymine (T), guanine (G), and cytosine (C). These nucleotides pair in specific configurations, forming the iconic double helix. But what if this alphabet could be expanded? The implications are compelling, ranging from personalized medicine to revolutionary materials.

“Life on Earth is amazingly diverse with just four nucleotides, so imagine what we could do with more,” said Dong Wang, Ph.D., a professor at Skaggs School of Pharmacy and Pharmaceutical Sciences at UC San Diego and the senior author of the study.

“By expanding the genetic code, we could create new molecules that have never been seen before and explore new ways of making proteins as therapeutics.”

Wang and his colleagues used a synthetic DNA system called AEGIS, which stands for Artificially Expanded Genetic Information System. AEGIS was created by Steven A. Benner, PhD, at the Foundation for Applied Molecular Evolution, as part of a NASA-funded project investigating how life could have evolved in other planets.

Dr. Dong Wang aptly describes that adding new ‘letters’ to the genetic code expands the vocabulary of life, allowing us to write more complex narratives.” His team’s breakthrough demonstrates that cells can readily incorporate synthetic nucleotides into the DNA recipe.

Using AEGIS

AEGIS adds two new letters to the standard DNA alphabet, which consists of adenine (A), thymine (T), guanine (G), and cytosine. These letters pair up in a specific way to form the double-helix structure of DNA, which was discovered by James Watson and Francis Crick in 1953.

The new letters, Z and P, have the same shape and size as the natural ones, so they can fit into the DNA helix without disrupting its geometry. This means that the enzymes that read and copy DNA, such as RNA polymerase, can recognize and process AEGIS DNA just like natural DNA.

The key lies in ‘mimicking’ nature’s machinery. The researchers identified RNA polymerase, a key enzyme that converts DNA into RNA, which is then used to make proteins. They designed two artificial nucleotides that flawlessly mimic the geometry of natural nucleotides. RNA polymerase readily accepted these novel additions when tested, seamlessly incorporating them into transcription.

The researchers tested whether RNA polymerase from bacteria could transcribe AEGIS DNA into RNA and found that it could do so with high accuracy and efficiency.

“This is a remarkable demonstration of how robust and adaptable the biological machinery is,” said Wang. “By mimicking the natural shape of DNA, our synthetic letters can sneak in and be used to make new proteins.”

This breakthrough paves the way for exciting possibilities. Imagine designing proteins with tailor-made properties capable of precisely targeting tumors for cancer therapy or engineering bacteria to synthesize eco-friendly biofuels. The vast horizons extend beyond medicine and environmental applications to materials science and potentially even synthetic biology.

Of course, challenges remain. Optimizing the incorporation of new nucleotides, ensuring their stability within the genome, and deciphering the full potential of this expanded code are areas for further exploration. Yet, the foundation for rewriting the genetic lexicon has been laid. This discovery signifies a momentous leap in our understanding of life’s blueprint. It holds the promise of ushering in a new era of biological design, where the possibilities are limited only by our imagination.  “These new proteins could have applications in medicine, biotechnology, and bioengineering,” said Wang. “We are only scratching the surface of what we can do with artificial DNA.”

Artificially Expanded Genetic Information Systems (AEGIS) add independently replicable unnatural nucleotide pairs to the natural G:C and A:T/U pairs found in native DNA, joining the unnatural pairs through alternative modes of hydrogen bonding. Whether and how AEGIS pairs are recognized and processed by multi-subunit cellular RNA polymerases (RNAPs) remains unknown. Here, we show that E. coli RNAP selectively recognizes unnatural nucleobases in a six-letter expanded genetic system.

 High-resolution cryo-EM structures of three RNAP elongation complexes containing template-substrate UBPs reveal the shared principles behind the recognition of AEGIS and natural base pairs. In these structures, RNAPs are captured in an active state, poised to perform the chemistry step. At this point, the unnatural base pair adopts a Watson-Crick geometry, and the trigger loop is folded into an active conformation, indicating that the mechanistic principles underlying recognition and incorporation of natural base pairs also apply to AEGIS unnatural base pairs.

These data validate the design philosophy of AEGIS unnatural basepairs. Further, we provide structural evidence supporting a long-standing hypothesis that pair mismatch during transcription occurs via tautomerization. Together, our work highlights the importance of Watson-Crick complementarity underlying the design principles of AEGIS base pair recognition.
